I know on Cyrene all the teleporters are allready available from start (if youtube video's can be trusted)... Is TP-run neccessary on Next Island to get them into the map (I wish the answer is yes)?
Yes, as far as I know you still need to do TP runs to get all the TPs here, and once you've done that you can then go to Ancient Greece and do runs for the TPs there as well.
